DNA Architecture: Nucleotides & Antiparallel Strands

Connect nucleotide chemistry to information storage and complementary pairing.

Safety, privacy and evidence boundary

Use paper/digital molecular models and fictional DNA sequences. No biological samples, genetic testing, ancestry inference, or personal genomic data are required. Molecular models are representations and should not be treated as photographs of atoms.

Explicit science instruction

DNA is a polymer of nucleotides, each containing deoxyribose, phosphate, and a nitrogenous base. Covalent phosphodiester bonds form a sugar-phosphate backbone. The two strands run in opposite 5′→3′ directions, making the double helix antiparallel.

Base pairing—A with T, G with C—allows sequence information on one strand to constrain the complementary strand while base stacking and backbone chemistry contribute to stability.

Vocabulary: nucleotide; phosphodiester bond; 5′ end; 3′ end; antiparallel; complementary; base pair; double helix.

Worked example with reasoning

Given 5′-A G T C C-3′, the complementary strand is 3′-T C A G G-5′. Written in its own 5′→3′ direction, it is 5′-G G A C T-3′.

Guided question

Why does strand direction matter when writing complementary DNA?

Independent work

  1. Label a nucleotide.
  2. Build a five-base antiparallel duplex.
  3. Write the complement both aligned and 5′→3′.
  4. Identify backbone versus information-bearing bases.

Misconception check

Hydrogen bonds between bases are not the covalent bonds that hold each DNA strand's backbone together.

Formative understanding check

Complement of 5′-ATGC-3′ written 5′→3′?

Worked response

5′-GCAT-3′.

Open-book formative practice only; not an official mastery attempt or grade.

Mentor evidence conference

Ask Archaemenes to challenge the model, quantitative reasoning, and limits without inferring personal genetic or medical information.

Open Archaemenes

Teacher guidance

Foundation: use color-coded nucleotide pieces. Core: track directionality precisely. Extended: connect chemical polarity to enzyme directionality and model limitations.

Preserve learning evidence

Keep the model, data interpretation, correction note, and limitation in the matching Week 23 evidence field.
